The supplement names three design principles: randomisation assigns treatments by chance; replication applies treatments to multiple independent experimental units to estimate variability; local control uses blocking or related restrictions to manage nuisance variation. An experimental unit is the smallest unit independently assigned a treatment. Repeated readings on one unit do not automatically provide independent replication. ANOVA may analyse an experiment, but the design and assumptions determine its valid form.
